Karakterisatie van neuromedine U-, tachykinine- en GnRH-verwante receptoren bij Caenorhabditis elegans

Signaaloverdracht tussen zenuwcellen: onderzoek op de rondworm C. elegans

Het belang van de vrijlevende rondworm Caenorhabditis elegans in wetenschappelijk onderzoek is niet te onderschatten, zoals blijkt uit de bekroning van zes C. elegans onderzoekers met maar liefst drie Nobelprijzen in de laatste elf jaar. Ook de onderzoeksgroep Functionele Genomics en Proteomics maakt dankbaar gebruik van de unieke eigenschappen van deze worm om meer inzicht te krijgen in de functies van neuropeptiden. Dit zijn kleine eiwitten die geproduceerd worden in het zenuwstelsel en zorgen voor de chemische communicatie tussen cellen. De structuur, signaalweg en uiteindelijke functie van deze neuropeptiden zijn vaak goed bewaard gebleven tijdens de evolutie. Hierdoor kan de kennis verzameld bij C. elegans bijdragen tot het beter begrijpen van gelijkaardige processen bij de mens en andere diersoorten.

Ook cellen kunnen met elkaar communicerenCommunicatie tussen cellen onderling en tussen cellen en hun omgeving is essentieel voor de regulatie van fundamentele biologische processen waaronder beweging, voortplanting, vertering, groei en ontwikkeling. Deze communicatie verloopt via signaalwegen waarbij een extern signaalmolecule, een ligand genaamd, wordt gedetecteerd door een specifieke receptor. De geactiveerde receptor geeft het signaal vervolgens door via boodschappermoleculen waardoor er een fysiologische respons wordt gegenereerd. Dergelijke signaalsystemen zijn van cruciaal belang voor het normaal functioneren van een organisme. Een verstoorde signaaltransductie kan namelijk aanleiding geven tot heel wat ziekten zoals kanker, diabetes en hart- en vaatziekten.

De neuropeptiden vormen de grootste klasse van signaalmoleculen bij meercellige dieren. Ze zijn onder meer betrokken bij de communicatie tussen hersencellen en hebben een sterke invloed op ons gedrag en gemoed. Het bestuderen van dergelijke signaalwegen in het menselijk brein is echter niet evident. Onze hersenen zijn een erg complex orgaan; ze bestaan uit vele miljarden zenuwcellen en ieder daarvan maakt gemiddeld contact met zo’n 10000 andere zenuwcellen. Daarom wordt voor het ontrafelen van neuropeptide signaaltransductie-systemen gewerkt met het modelorganisme C. elegans. Net zoals de mens, maakt ook deze kleine bodemworm gebruik van neuropeptiden voor de intercellulaire communicatie. Maar in tegenstelling tot de miljarden zenuwen in het menselijk brein, beschikt een volwassen worm over slechts 302 zenuwcellen. Dit kan erg eenvoudig lijken, maar desondanks zijn de biochemische signaalprocessen verrassend gelijkend aan deze in meer complexe organismen, inclusief die bij de mens.

Identificatie van nieuwe neuropeptide-receptor koppelsVolgens computersimulaties behoort 7% van de eiwit-coderende genen in C. elegans tot de familie van G-eiwit gekoppelde receptoren (GPCR’s). Deze receptoren danken hun naam aan de zogenaamde G-eiwitten. Dit zijn boodschappermoleculen die door een GPCR geactiveerd worden, het signaal doorgeven in de cel en zo een verandering in de cel bewerkstelligen. De GPCR’s vormen bovendien de belangrijkste groep van moleculaire doelwitmoleculen voor therapeutische geneesmiddelen: ongeveer 40% van de huidige medicijnen zijn gericht tegen GPCR’s. Ondanks deze voorspellingen zijn er nog een groot aantal C. elegans GPCR’s waarvan het activerende ligand niet gekend is, de zogenaamde weesreceptoren.

In deze masterthesis werd er op zoek gegaan naar de liganden van een aantal weesreceptoren. We gebruikten hiervoor een techniek die bekend staat als ‘omgekeerde farmacologie’, waarbij de receptor dienst doet als lokaas om de activerende signaalmoleculen op te vissen. Bij het uitvoeren van zo’n experiment wordt de weesreceptor in contact gebracht met 261 verschillende synthetische C. elegans peptiden. Wanneer een peptide erin slaagt om aan de receptor te binden en deze te activeren, wordt er een fluorescent lichtsignaal geproduceerd dat wordt opgevangen door een detector. Op deze manier zijn we erin geslaagd om twee voorspelde C. elegans GPCR’s te ‘ontwezen’. De eerste receptor (CE16937) wordt geactiveerd door twee peptiden die verwant zijn met de tachykininen. De tachykininen vormen een multifunctionele peptidenfamilie en komen zowel bij ongewervelde als gewervelde diersoorten voor. Het bekendste lid van de tachykininenfamilie is ‘Substantie P’, een neuropeptide dat onder meer betrokken is bij het waarnemen van pijn. Het tweede ontdekte neuropeptide-receptor signaalsysteem is verwant met de neuromedine U signaalweg bij de gewervelde dieren. Leden van de neuromedine U peptidenfamilie vervullen uiteenlopende biologische functies, gaande van het stimuleren van spiercontracties tot het reguleren van stress.

Lokalisatie en functionele karakterisatie van een tachykinine-verwant signaalsysteemVervolgens legden we ons toe op een verdere karakterisering van het nieuw ontdekte tachykinine-verwant signaalsysteem met als doel de functie ervan te ontrafelen in de worm. Hiervoor zijn we nagegaan in welke cellen de receptor (CE16937) en de twee tachykinine peptiden tot expressie komen. Wetenschappers hebben hiervoor een erg handig trucje uitgevonden. Door het eiwit van interesse, in dit geval de receptor of de neuropeptiden, te koppelen aan een groen fluorescent proteïne (GFP), wordt het visueel waarneembaar onder een gewone microscoop uitgerust met een fluorescente lichtbron. En doordat C. elegans transparant is, is het zelfs mogelijk om deze fusie-eiwitten in een levende worm te observeren. Het tachykininepeptide-GFP fusie-eiwit werd waargenomen in een aantal zenuwcellen in de kop en de staart van de worm, alsook in spiercellen van de lichaamswand. Verder onderzoek is nodig om deze zenuwcellen te identificeren en na te gaan waar de receptor gelokaliseerd is. Kennis over de cellulaire lokalisatie van de receptor en zijn liganden kan namelijk aanwijzingen geven over de functionele rol van het neuropeptide signaalsysteem aangezien de positie van alle 959 cellen, alsook de functie van veel van deze cellen, in detail gekend is.

Heel wat ziektegenen en signaalwegen betrokken bij aandoeningen bij de mens blijken ook aanwezig te zijn bij de nematode C. elegans. Deze conservering alsook de beschikbaarheid van een uitgebreide genetische toolbox maakt van de worm een ideaal modelorganisme voor het ontrafelen van dergelijke communicatiemechanismen. De identificatie van twee nieuwe C. elegans neuropeptide signaalsystemen is een eerste stap vooruit om betere inzichten te verwerven in de moleculaire werkingsmechanismen en de functies van deze complexe systemen.
