High-throughput screening of antimicrobial peptides against Candida krusei and Cryptococcus neoformans

Het optreden van resistentie tegen het huidige antibioticagamma bij verscheidene micro-organismen resulteert in een expanderende gezondheidsgerelateerde bedreiging op wereldwijd niveau. In de zoektocht naar vervangers voor de falende klassieke antibioticatherapie worden antimicrobiële peptiden beschouwd als één van de veelbelovende alternatieven. Voor het onderzoek “High-throughput screening of  antimicrobial peptides against Candida krusei and Cryptococcus neoformans” werden twee peptide databanken getest op hun antimicrobiële activiteit tegen de fungi Candida krusei en Cryptococcus neoformans. Hieruit werden enkele veelbelovende kandidaten gedetecteerd die in verder onderzoek en na eventuele optimalisatiestappen als mogelijke substitutietherapie van de actuele antifungische agentia kunnen aangewend worden.

Antimicrobiële peptiden als alternatieven voor de huidige resistentieonderhevige antibiotica

Antimicrobiële peptiden zijn beloftevolle alternatieven in de zoektocht naar nieuwe potentiële geneesmiddelen tegen micro-organismen, die ten gevolge van het huidige, toenemend probleem van (multi)drugresistentie een risico voor de humane gezondheid vormen op een universeel niveau. Deze in verscheidene species natuurlijke voorkomende peptiden vertonen een breed spectrum activiteit, een snelle werking en een lage vatbaarheid voor resistentie. Ze worden mede daardoor onderzocht tegen verscheidene micro-organismen, waaronder fungi. Fungi zijn één van de primaire oorzaken van letale infecties in immunogecompromitteerde patiënten. De huidige bestaande antifungische geneesmiddelen zijn gelimiteerd, niet zo efficiënt als gewenst en zijn ook onderworpen aan resistentie problemen.

Antimicrobiële activiteit van de peptide databanken testen via een op fluorescentie gebaseerde screening methode

Het hoofdobjectief van het onderzoek was het identificeren van veelbelovende antimicrobiële peptiden actief tegen de fungi Candida krusei en Cryptococcus neoformans via een op fluorescentie gebaseerde high-troughput screening van twee peptidenbibliotheken. Deze screening methode is gebaseerd op de blauwe kleurstof resazurine, die een lage intrinsieke fluorescentie heeft en door viabele fungale cellen tot de roze fluorescerende resorufin gereduceerd wordt. De gemeten fluorescentie correleert met de hoeveelheid viabele cellen en daardoor met de activiteit van de peptiden. Het antifungisch potentieel van 240 synthetische nonapeptiden en van 430 in de natuur voorkomende peptiden werd onderzocht.

De beloftevolle kandidaten

Zeven sterk actieve peptiden werden gevonden tegen Cryptococcus neoformans, waarvan twee van de synthetische peptidenbibliotheek en vijf van de natuurlijke peptidenbibliotheek afkomstig zijn. Negen peptiden van de natuurlijke peptide bibliotheek werden zeer actief bevonden tegen Candida krusei, terwijl geen synthetische peptiden sterke activiteit tegen dit micro-organisme vertoonden. Zowel selectieve peptiden tegen Candida krusei alleen (één synthetische peptide en drie natuurlijke peptiden) of tegen Cryptococcus neoformans alleen (één synthetische peptide en één natuurlijke peptide) als breed spectrum peptiden (twee synthetische peptiden en vijf natuurlijke peptiden) werden gedetecteerd.

Correlatie tussen antimicrobiële activiteit en karakteristieken van de peptiden

Daarbovenop was het doel om informatie over belangrijke karakteristieken van de peptiden te bekomen die helpen om  de peptiden te onderscheiden op basis van hun activiteit. Een analyse van de distributie en de posities van de aminozuren in de peptidensequenties en een eendimensionale ANOVA analyse betreffende deze peptide eigenschappen worden hiervoor uitgevoerd. Algemeen kon besloten worden dat de juiste balans tussen hydrofobe en positief geladen aminozuren vereist is om sterk actieve peptiden te verkrijgen tegen de geteste fungi.

Toekomstig werk

De interessant bevonden peptiden kunnen geoptimaliseerd worden op basis van voorgaande analyse resultaten en andere technieken. Ze kunnen dan opnieuw gescreend worden tot het gewenste therapeutische potentieel bereikt is.
