Ontwikkeling van een expressiesysteem voor de productie van het "Ice Nucleation Protein"

Bacteriën laten water sneller bevriezen

Bevriezen van water is duur

In tegenstelling tot wat de meeste mensen denken, bevriest water niet bij 0°C. Het water kan echter afgekoeld worden tot ver onder het nulpunt vooraleer er effectief ijs gevormd wordt. Zo kunnen kleine volumes ultrazuiver water afgekoeld worden tot een temperatuur van -40°C voordat bevriezing optreedt. Enkel bij oneffenheden in het recipiënt of aanwezigheid van onzuiverheden of vreemde partikels in het water, kan het water bevriezen bij een hogere temperatuur. In de meeste installaties moet het water afgekoeld worden tot ongeveer -10°C om ijsvorming te bekomen.  Doordat water niet bevriest op het nulpunt heeft dit een nadelig effect op toepassingen waarbij ijs gevormd moet worden. Hierdoor is er bijvoorbeeld veel koelingsenergie nodig om kunstsneeuw aan te maken of een ijspiste aan te leggen.

Wanneer er aan water onzuiverheden worden toegevoegd, kan de benodigde koeling gereduceerd worden. Zo voegde men vroeger mineraalzouten, zoals zilverjodide, toe aan het water om goedkoper sneeuw te kunnen maken. Dit was mogelijk doordat deze zouten een kristalstructuur hebben die zeer sterk gelijkt op die van ijskristallen. Hierdoor kunnen de watermoleculen zich afzetten op de zoutkristallen waardoor het water makkelijker bevriest. Dit zorgt ervoor dat het water afgekoeld moet worden tot slechts -8°C. Omdat veelvuldig gebruik van dergelijke mineraalzouten een negatieve invloed heeft op mens en natuur, werd het gebruik er van snel opgeschort en is er dus nood aan een duurzaam alternatief om ijsvorming te versnellen. 

Bacterieel proteïne als ijsmaker

Nu blijkt dat er verschillende soorten bacteriën zijn die een eiwit kunnen aanmaken dat water bij een hogere temperatuur kan laten bevriezen. Deze bacteriën komen in de natuur voor op de bladeren van verschillende planten. Het bezit van dit eiwit biedt de bacteriën enkele voordelen. Onder normale omstandigheden kunnen vorstgevoelige planten tegen temperaturen onder het nulpunt. Echter wanneer nu deze bacteriën op die planten aanwezig zijn, kan dit vorstschade veroorzaken. Deze vorstschade is het gevolg van het scheuren van plantencellen door de vorming van het ijs, waardoor de inhoud van de plantencel vrijkomt en als voedingsbron gebruikt kan worden door de bacterie.

Deze bacteriën produceren het zogenaamde Ice Nucleation Protein (INP). Door dit eiwit kunnen de bacteriën water bevriezen bij temperaturen van slechts -2,5°C. Een eiwit bestaat uit een aaneenschakeling van verschillende bouwstenen, die men aminozuren noemt. Elk eiwit bestaat uit een unieke aaneenschakeling van deze aminozuren. Uit onderzoek is gebleken dat de structuur en de opeenvolging van de bouwstenen (aminozuren) van het Ice Nucleation Proteïne, afkomstig van verschillende bacteriën, zeer sterk op elkaar gelijken. Bovendien hebben alle teruggevonden INPs een centraal domein dat bestaat uit een bepaald aminozuurpatroon, bestaande uit 8 aminozuren dat een 30-tal keer achter elkaar herhaald wordt. Dit geeft aan dat deze opeenvolging zeer belangrijk is om het water te kunnen laten bevriezen. Zo zou het eiwit dienst doen als een sjabloon om watermoleculen te ordenen in een ijskristal.  

Productie uitbesteden aan andere bacterie

Het is duidelijk dat het gebruik van bacteriën die een INP aanmaken, voordelen kan bieden ten opzichte van mineraalzouten in tal van processen waarin ijsvorming beoogd wordt. Ten eerste kunnen ze het water bevriezen bij hogere temperaturen. Dit zou kunnen zorgen voor verdere besparing van energie nodig voor koeling. Ten tweede zijn deze eiwitten natuurlijke producten en onschadelijk voor mens en natuur. Echter niet elke bacterie kan met behulp van zijn INP het water bevriezen bij eenzelfde temperatuur. Daarom werd in een eerste fase van dit onderzoek een 100-tal verschillende bacteriën gescreend op de aanwezigheid van een gen dat codeert voor het INP. Vervolgens werd nagegaan bij welke temperatuur deze bacteriën ijsvorming kunnen induceren.

In natuurlijke omstandigheden wordt het INP echter aangemaakt in relatief beperkte hoeveelheden. Om dit INP in te kunnen zetten in industriële toepassingen, is bijgevolg een hogere productie nodig. In een tweede fase van dit onderzoek werd verder gewerkt met een bacterie die bij de hoogste temperatuur ijsvorming kan induceren en werd de genetische informatie die codeert voor het INP lichtjes aangepast en in een andere bacterie gebracht.

Op deze manier kan de productie van het gewenste Ice Nucleation Protein uitgevoerd worden door een andere bacterie, Escherichia coli (E. coli). Deze darmbacterie wordt veel gebruikt voor de productie van zogenaamde recombinante eiwitten omdat E. coli makkelijk manipuleerbaar is, genetisch goed gekend is, snel groeit en groeit op goedkope grondstoffen. Hierdoor is de opgroei van deze bacterie en dus ook de productie van het INP goedkoper.

In dit onderzoek zijn we er dus in geslaagd om een nieuw systeem te ontwikkelen waardoor E. coli in staat is om het Ice Nucleation Protein te produceren in grotere hoeveelheden.  Hierdoor vertoont deze gemanipuleerde E. coli zelfs een hogere activiteit dan de natuurlijke organismen en kan het water bevriezen bij een temperatuur van-2,5°C. Door de hogere activiteit zijn er bovendien minder bacteriën nodig om éénzelfde volume water te bevriezen.

Op basis van deze nieuwe, gemanipuleerde bacterie is het dus mogelijk om nieuwe, efficiëntere producten aan te maken om water goedkoper en sneller te laten bevriezen. Daarenboven kan via dit nieuwe productiesysteem het eiwit zelf snel en goedkoop opgezuiverd worden, waardoor het afzonderlijk gebruikt kan worden in plaats van de bacteriën. Gezien het gebruik van genetisch gemodificeerde organismen (GGO) strikt gereglementeerd is en de publieke opinie eerder terughoudend is rond het gebruik van GGO’s, biedt een opgezuiverd proteïne meer mogelijkheden voor toepassingen. Zo kan het INP in de toekomst mogelijk gebruikt worden om voedingswaren of koelelementen sneller, goedkoper en duurzamer in te vriezen. Daarnaast heeft het een groot potentieel om ijspistes aan te leggen met minder verbruik van (koelings)energie of het kan ook gebruikt worden in sneeuwkanonnen. Hierdoor is het mogelijk dat wanneer je nog eens gaat skiën of schaatsen je dit op sneeuw of ijs doet dat werd aangemaakt met de hulp van deze bacteriën.
