Planten vaccineren?

Vaccinaties, we kunnen er in deze periode niet omheen. Een vaccin stimuleert het immuunsysteem, zodat we gepaster kunnen reageren op een infectie. Op dit moment worden vaccinaties als een van de uitwegen gezien uit de SARS-CoV-2 pandemie. Waar echter minder mensen van op de hoogte zijn, is dat planten ook een immuunsysteem bezitten dat gestimuleerd kan worden, waardoor ze zich beter kunnen weren tegen ziektes en stressfactoren. Hierdoor kan men opbrengstverliezen veroorzaakt door deze stressfactoren verlagen. Het vaccineren van planten noemt men “geïnduceerde resistentie”. In combinatie met andere technieken om ziekten bij planten te voorkomen, zou dit een belangrijke rol kunnen spelen bij de verhoging van landbouwopbrengsten, een noodzaak om de groeiende wereldbevolking te kunnen voeden.

‘Geen honger’, zo luidt de tweede doelstelling van de Duurzame Ontwikkelingsdoelstellingen, vastgelegd door de Verenigde Naties. Hierbij streeft men, via een duurzaam landbouwsysteem, naar de uitroeiing van honger en het garanderen van voedselzekerheid tegen 2030. Verschillende uitdagingen bedreigen echter het behalen van deze doelstelling. Ten eerste groeit de wereldbevolking, terwijl de beschikbare landbouwgrond niet evenredig kan worden uitgebreid. Ten tweede bedreigen stressfactoren de opbrengst van landbouwgewassen. Deze factoren kunnen enerzijds afkomstig zijn van biotische of levende bronnen, zoals schimmels, insecten en nematoden. Anderzijds kunnen stressfactoren abiotisch zijn, uit de niet levende natuur, droogte of warmte bijvoorbeeld. Om gewasopbrengsten op een duurzame manier te verhogen, worden traditionele technologieën, zoals veredeling en gewasbeschermingsmiddelen, gecombineerd met nieuwere, modernere technieken. Een van deze moderne technieken is het stimuleren van het immuunsysteem van planten, zodat zij beter opgewassen zijn  tegen stressfactoren.

Het plantenimmuunsysteem

Planten kunnen zich niet voortbewegen en hebben daarom een vernuftig immuunsysteem ontwikkeld waarmee ze kunnen reageren op stressfactoren in hun nabijheid. Een plant gebruikt receptoren, specifieke elementen of eiwitstructuren, om patronen uit de omgeving te herkennen. Als de plant een patroon herkent, kan hij een signaal zenden naar de celkern. Die celkern is een soort van bibliotheek, die alle informatie van de plantencel – het zogenaamde DNA – bevat. Het signaal zal ervoor zorgen dat een specifiek deel – zoals een boek – wordt gelezen en overgeschreven. Het boek zelf mag de bibliotheek niet verlaten, anders zou het schade kunnen oplopen of verloren raken. Het overgeschreven deel – het zogenaamde mRNA – mag de kern wel verlaten, en gaat naar de werkplaats van de cel, waar gepast gereageerd wordt op het signaal van buitenaf. Zo kan hij ervoor zorgen dat goedaardige bacteriën, die hem een voordeel leveren, in zijn buurt kunnen blijven. Anderzijds kan de plant proberen stressfactoren te elimineren. Hij kan bijvoorbeeld stoffen aanmaken waardoor hij beter bestand is tegen stressfactoren, of stoffen die schadelijk zijn voor ziekteverwekkers. Hij kan er ook voor zorgen dat bepaalde van zijn cellen sterven, zodat de ziekteverwekkers zich niet verder verspreiden.

Bij het verkrijgen van geïnduceerde resistentie geeft men een stimulus aan de plant, waardoor zijn verdedigingscapaciteit verbetert. Deze stimulus kan een chemische stof zijn, een goedaardige bacterie, een goedaardige schimmel of een ziekteverwekker. Wanneer de plant in contact komt met de stimulus, maken verschillende veranderingen een verbetering van de reactie mogelijk, waarna hij sneller en gevatter kan reageren op bepaalde stressfactoren.  Ten eerste kan hij meer receptoren maken, die patronen uit de omgeving kunnen herkennen. Ten tweede kan hij meer inactieve signalen aanmaken, als een soort van reservevoorraad. Bij het herkennen van een patroon kan de plant deze signalen activeren, waardoor ze sneller naar de kern worden doorgestuurd. Tot slot is het mogelijk dat de plant de structuur van zijn bibliotheek, waar al het DNA zich in bevindt, aanpast. Hierdoor kan de plant sneller of makkelijker bepaalde informatie overschrijven. Dankzij deze aanpassingen kan de plant sneller reageren op signalen van buitenaf, de gewonnen tijd kan cruciaal zijn om de ziekteverwekker te bestrijden. Afhankelijk van de plantensoort en de stressconditie reageren planten anders op een bepaalde stimulus. Het is dan ook belangrijk om altijd na te gaan of een stimulus wel helpt om het immuunsysteem te verbeteren tegen een bepaalde stressconditie in een bepaalde plantensoort.

Zuur en bacteriën 

Er zijn al producten op de markt die planten beschermen tegen ziektes door hun immuunsysteem te stimuleren. Toch blijft verder onderzoek naar nieuwe stimuli nodig. Een van die nieuwe stimulerende producten is piperonylzuur, een chemische stof. Voorafgaand onderzoek had aangetoond dat piperonylzuur het immuunsysteem van rijst stimuleert tegen het wortelknobbelaaltje, een nematode. In mijn onderzoek wilden we nagaan of piperonylzuur rijst eveneens kan beschermen tegen een schimmel, “Pyricularia oryzae”. Deze schimmel infecteert de plantencellen, laat ze even leven om voedingstoffen te gebruiken, maar doodt ze uiteindelijk toch. Hierdoor kan deze schimmel een belangrijke oorzaak zijn van oogstverliezen. Wanneer planten vooraf een behandeling met piperonylzuur ondergingen, vertoonden zij minder symptomen veroorzaakt door de schimmel. Dit is een aanwijzing dat het immuunsysteem van de plant beter kon reageren op de schimmel dankzij de voorgaande behandeling met piperonylzuur.

In een tweede reeks experimenten werd het effect van stoffen geproduceerd door goedaardige bacteriën, “Pseudomonas” bacteriën, op nematoden onderzocht. Pseudomonas bacteriën zijn divers, en ze produceren verschillende soorten stoffen. Eerder onderzoek had al aangetoond dat enkele van deze bacteriën het immuunsysteem van rijst stimuleert tegen de hierboven vermelde schimmel. We toonden aan dat een of meerdere stoffen die deze bacteriën produceerden ervoor zorgden dat nematoden minder actief worden. Er kan echter niet met zekerheid gezegd worden welke van deze geproduceerde stoffen dit effect hebben op de nematoden.

Beter immuunsysteem, hogere opbrengsten 

In mijn thesis toonden we de mogelijkheden van de stimulatie van het plantenimmuunsysteem verder aan en twee soorten stimuli werden verder gekarakteriseerd. Dankzij deze stimulatie zijn planten weerbaarder tegen ziekten en plagen. Dit kan leiden tot minder opbrengstverliezen in de landbouw en een stabielere voedselketen, waarbij minder gewasbeschermingsmiddelen gebruikt moeten worden. Op deze manier kunnen we verder bouwen aan een meer duurzame landbouw.
